After VISTA SP1 has been automatically installed on my computer is there any
way I can retrieve the file again from my HD and burn it to a CD for use on
my wife's computer?

Probably not. If instead you would have went to the Microsoft site and
just download the executable SP1 file then yes since at that point it
is such another file. Which of the many components gets installed
depends on the system it is installed on. So what your system might
have needed and got changed may be very different that what your
wife's computer might have benefited from.

You should have downloaded the standalone package that has been
designed to allow installations on more than one computer. Even if you
only have on computer, the standalone package is the better choice,
because if for whatever reason you re-install Windows Vista, you'll not
have to download the SP1 again.
