desktop not operating with keyboard or mouse

Help! I cannot access my desktop from the start menu, with mouse or keyboard.
I have made some changes on pc and since get this pop up saying: could not
load or run lllllll lllllllllllllll specified in the registry, However if I
leave the pop up on the desk top with out closing it, I can then use and
access all programes, files and internet access. If I close it, nothing
works. I an using xp home os. Any help would be appreciated.
