Administrator is treated in Vista very different than in XP and previous.
Due to UserAccountControl /"UAC" you are logged on as "a local admin", but
not as "root", meaning "admin with systemwide access. Whenever you want to
perform action that needs root access, you will be presented the UAC shield
where Vista asks for your OK.
This was intended to prevent Malware ffrom self-installing without your ok
and so on, the safe Desktop ( grey Desktop ) is to prevent Malware from
"self-clicking the ok button"..
To perform file-actions in the Windir and subdirectories like /system32 you
will not only need root-access, but need to take ownership of each
individual file. ( IN XP the files were owned by You,the user - in Vista
critival are owned by the OS. )
Get used to this new behaviour, it is for the greater good and is the
critical point behind Vistas security policy.
If you cannot for what reason ever, there is the possibility to switch back
to XP behaviour, by a) turning off UAC b) enable the "root" Admin fully and
make use of the "takeown" command in said directories.
you might have the need to use the comands gpedit.msc and secpol.msc to make
changes to the way the Admin-Account is treated. Note that these Commands
are not in the Home-Editions of Vista.

I recommand a special file-manager which has built in "do this as the Admin"
capabilities :

maintained by Christian Ghisler :

Clone, but with so much more... To use the Windows Explorer for such stuff
is really not worth it.
