First, I'm not an expert here but you could WinZip that folder (or the
contents of that folder) and give it a password (from WinZip). That might
not be as elegant as you would want but it would do the trick.

Hi,

Natively? No, Vista uses permission sets to determine access to a
file/folder. Passwording is inherently inferior. There are third party
applications, most commonly associated with zipping the file, that will do
this.

No, you cannot, but you can use EFS (Encrypting Files System).
File/Folder Properties, Advanced, Encrypt Contents...

But, before you start using EFS, you should learn how it works.
You MUST save your EFS certificate, or you risk loosing your encrypted data.

Not natively in some versions of Vist
