How do I set up proxy access to another users mailbox

Seceratry needs access to Boss's email from her desktop

Exchange version? SBS version?

Different rules for differen versions, and your details are
non-existent.

Ok, Active Directory users and Computers (or start, run, dsa.msc)

Find the user, go to their properties, then click on the Exchange
Advanced tab

Click Mailbox Rights

Add the user who you wish to grant access to this mailbox. Tick all
the boxes down to "take ownership".

The user can now open this mailbox within outlook.

Thanks - I've done that but I only get an option to Open a Folder such as
Inbox

No option to open a "Mailbox" an add it to the Tree on the left

I find the easiest way here is to go into the user's account
properties in Outlook, where you can choose to "open additional
mailboxes". Is that what you did, or somethign different?
