Hello The,
My default Windows Server 2008 R2 server install is using roundabout 11GB
diskspace until now with the latest updates. Windows Server 2008 with SP2
and the latest updates uses roundabout 10 GB. So i think even with future
SPs you will not reach 72 or 100 GB disk space.
Check this article about AD database file changes/problems within Windows
server 2008/2008 R2:

More important that using different harddisks in my opinion is that you use
a tl east a mirrored drive in case one disk fails. Also it is recommended
that you store the Active Directory database and the SYSVOL shared folder
on the same drive.
Best regards
